NHR2-inhibitors - a new approach to the treatment of leukemia
Acute myeloid leukemia (AML) is one of the most frequent cancers of white blood cells. An important event in the course of the disease is the aggregation of certain proteins to groups of four (tetramers). These proteins are named RUNX1/ETO, which regulate the expression of several genes in white blood cells.
The inventors have identified a new class of substances that inhibit the aggre-gation of the proteins to tetramers. Therefore, these substances appear eligible for the treatment of this type of cancer. The efficancy of several of these sub-stances has been demonstrated in isolated cells as well as by in vivo models, whereas the latter has shown a significant reduction of tumor growth.
Property rights have been enrolled in the USA and Europe for those substances that are promising for the treatment of AML.

NMDAR antagonists as novel anti-diabetic drugs
The N-Methyl-D-Aspartat (NMDA)-receptor is well known as receptor of the nervous system. The substance Dextromethorphan is an antagonist for the NMDA-receptor and a long-standing drug for the repression of tussive irritation and other applications.
Scientists of the Heinrich Heine University of Düsseldorf have recently discov-ered that the NMDA-receptor is also present on cells of the pancreas and has influence of the insulin-production. Therefore, antagonists for this receptor such as Dextromethorphan can be used as novel anti-diabetic drugs. A significant advantage in comparison to other anti-diabetic drugs is the mode of action: the basal insulin-release is not affected, which consequently minimizes the risk of hypoglycemia.
In addition, the scientists have generated new Dextromethorphan-derivatives, which have significantly less unwanted side-effects on the nervous system than the original substance. A further advantage of the new substances is their ap-parent protective effect on the insulin-producing beta-cells of the pancreas, which slows down the progression of the diabetes. Initial clinical studies have provided evidence that Dextromethorphan has a good synergistic effect in com-bination with other anti-diabetic drugs.
The substances and their use as anti-diabetic drugs have been enrolled for pa-tent in Europe.

Inhibitors for the AKT-enzyme with a novel, covalent-allosteric mode of binding
The inhibitor Herceptin was the first representative of the so-called „second wave agents“, which is a new class of anti-cancer drugs that is characterized by the selection of specific target molecules with known involvement in cellular events of the cancer disease. The enzyme AKT is such a molecule, which plays an important role in the survival of the tumor cells in several types of cancer.
Researcher of the Technical University of Dortmund have developed inhibitors for AKT with an entirely new mode of binding. This special covalent-allosteric mode of binding provokes an irreversible and specific inhibition of the AKT at a very low drug concentration. This is a decisive advantage in comparison to ex-isting AKT inhibitors. The scientists are currently working on the confirmation of efficacy of their substances in an in vivo model.
The substances and their use as anti-cancer drugs have been enrolled for patent in Europe.
